Speaker of the house is entirely different from the President of the Senate. Speaker of the House is the Head of the House of the Representative. Member of the House of Representative are called as CONGRESSMEN. In the Senate, it is headed by the Senate President and the elected Senate members are called SENATORS. In the event of vacancy in the Vice Presidents Office, the President of the Senate automatically assume the Vice Presidency of the country and the speaker pro tempore ascended to the senate presidency.

The President of the Senate is actually the Vice-President of the United States. I don't think it would be necessary for the Speaker to act in his absence, unless there was a tie vote, which is the only time the President of the Senate votes.

The Speaker of the House is from the House of Representatives instead of the Senate and acts as the preceding officer of the House. If something were to happen to the President and the Vice-President, the Speaker of the House would run the country.
