It depends on your relationship with your brother, as much as it does their relationship.
First: your brother is not trying to eliminate a rival for your affection (obviously!) so you don't have to worry about that as a motive. Then again, if he doesn't like your boyfriend, he certainly has a potential motive for lying.
But, but, but...why would he bother? Let's say he doesn't like the guy for reasons that have nothing to do with you. It's not like he has to date your boyfriend; he doesn't need to have anything to do with him. So it's certainly possible that your brother knows something about your boyfriend, or sees something in his behavior, that has him concerned for your well-being -- and that's why they don't get along.
On the other hand, people with low expectations may not interpret things accurately. Once upon a time, an ex-boyfriend of my new girlfriend ran into me, and said "So, you're taking x to the prom?" and I said "Looks that way." He went to her later that day, and said I had replied "Unfortunately." It's entirely possible he misheard, and assumed the worst. Your brother might be doing the same.
Still, at the end of the day, if your brother and you get along, he's probably looking out for your best interests; his heart is probably in the right place. If this thing that your boyfriend supposedly said is bad enough to destroy your relationship, I'd confront your boyfriend and see how he reacts. Ideally, you'll do it in a "ha ha, did you really say that?" way, and perhaps he'll admit it (thinking it doesn't really bother you.)
The thing is, if your relationship is already on such thin ice that a single statement can destroy it, you're on the brink of breaking up anyway (unless the thing he said was something like "I'm gay" or "I cheated on her, so what?") -- so keep that in mind, and don't trust your brother OR your boyfriend: trust yourself and your gut.

It depends on what it is about. If they dislike each other enough then they may try to pull you away from the other, so if they have shown any type of spitefull characteristic i the past, then I would take what is being said with a grain of salt. Now if they have been honest and fair in the past i think I would still believe my brother before my boyfriend. But I am very close with my brother so it is all dependent upon ur relationship

Well you should always believe your family for one in most cases but if your brother has always hated your boyfriend because of whatever reason he might just be lying so that you will get mad at him and break-up with him. Also you have known your brother for however long maybe you should sit down and talk with him just the two of you because you should beable to tell if he is lying or not.

they say blood is thicker than water,put both face to face and ask the question,if your boyfriend starts squirming believe your brother,boyfriends have a habit of telling things and lying about it,but if you know what brother said originally and it changes,believe boyfriend,means brother trying to set up boyfriend.

I would believe your brother. He is family and has no reason to lie to you. If he is telling you something bad your boyfriend said..it's probably true. As your brother is probably trying to protect you from being hurt and used by your boyfriend. And if you're close to your brother he'll try to protect you.

Family only if your brother is not the kind of person who would lie to make you stay away from the opposite sex.By him being your brother a lot of brothers will tell their sisters things that they need to to know even though it may hurt them.Trust me it is classic case scenario.

Umm.. if it's something that is between you and your boyfriend that no one knows, and your brother knows, I would believe your brother. Of course, if it's something that your brother could guess at, and you had a reaction to it, you told him yourself... I would trust your brother...
