Hmmm. Not sure what you mean to express.

"I don't know why do you think" Do you mean "I don't know why you think [that the Earth is flat, when clearly it's round]" or "I don't know why. Why do you think?"

"I am not sure do you think" could mean "I am not sure if you think [that I like you, but I don't]" or "I am not sure what you think" or "I am not sure. Do you think [we need to bring a sweater]?"

"I am glad with you" could be "I am glad, too" (you're glad and so am I) or "I am glad for you" (whatever you received or achieved that was good, makes me glad).

the 3rd phrase: i am glad too.
