I assume that you do not want a religious work. Since you require a work of fiction, that rules out Homer's "The Iliad," which is epic poetry. It also rules out everything by the rest of the Greeks and all of the Romans. This also rules out a lot of work by the Chinese, Japanese, Arabs, etc.
Thus, I might suggest Thomas Malory's Le Morte d'Arthur, which may not be the first novel written (there are many candidates), but it might be the first work of fiction. The nature of your question could demand the origin of fiction for an answer.
But who reads Le Morte d'Arthur? What's a book of fiction that EVERYONE reads at some point in their lives? I can't think of any more widely read than Mary Shelley's Frankenstein.
But does originality and popularity necessitate "greatest impact?" If not Le Morte d'Arthur and not Frankenstein, perhaps, then, George Orwell's 1984.

i think that brave new world and the giver impacted people about what life could be if they try to make a utopia both are fiction and they are really good, but that's just me, i like almost every book i read, but i'm picky about books

one person mentioned brave new world and the giver, well, a handmaid's tale falls under that category also. i liked it better than brave new world, i loved the giver. but a handmaid's tale is worth mentioning also. the jungle is a great example also because it forced the government to start looking into meat packing industry and develop regulations regarding food.
