well--I think you first can be a little bit flattered, I mean someone finding you attractive is always a positive thing! :-) then just be polite and let her down easy like you would if a guy you weren't digging hit on you!
It's not a huge deal, in my very limited experience the girls that have hit on me were either a) very, very drunk or b) knew that I was straight and were just messing around.

I assume your response stems from the fact that you're straight, correct? If that's the case, I would handle this very tactfully. Given all the sickening homophobia that exists in this country think for a moment how incredibly difficult it was for this woman to make a pass at you. Not only did she risk being rejected because you're not romantically attracted to her (bad enough) but she also risks being humiliated, ostrascized (despite the mounthbreathing frat boy porn mentality that runs rampant in this country), and generally having her life ruined because of it.
My very serious advice is to first, take a deep breath. Second, consider what an enormous compliment this woman paid you given the risks she's taking (and let's be honest, guys will hit on a first cousin in a pinch). Third, gracefully and as nicely as you can, tell this woman that as flattered and surprised as you are (how many people are attractive to both genders?), you are comfortably and happily straight.
Remember, you have a great deal of responsibilty in this situation and how you handle it says a lot about you as a person.
