I think Christ was referring to the idea that the Spirit is invisible, inscrutable and incomprehensible. In other words, it is something beyond the realm of empirical evidence and knowledge. Science can neither confirm nor deny it's existence or function.

We come and go unseen. When the wind blows, can you tell if it is coming or going? Can you look at a stranger who is walking down the street and say "That is a christian." if they have no outward sign of their faith? We are like the wind in the sense that we carry the world of the Lord which he likened like a seed in one of his parables. There are many explainations for it.
If you are born of the spirit it means you have the Holy Spirit within you. You are reborn, or born again.

Because when you are born again you are given the holy spirit. And when you have the holy spirit, you hav e the power to do anything cause you become a child of god.
