This is a good website about Harlem's various neighborhoods: http://www.myharlem.org/v2/index.php
Columbia, Barnard and City College are all located in Harlem. I'm not particularly familiar with City College, but the neighborhood surrounding Columbia and Barnard (they're across the street from each other) is fairly safe. You need to take the 1 subway line to all of these schools. Columbia and Barnard are at 116th street and City College is at the 137th street stop. Buses also make stops at these schools, but I don't use them.
Regarding diversity, I assume you mean on campus. Columbia is definitely VERY white. However, there is quite a large Asian community and all others are well-represented. This is not to say that I haven't had classes where there was not one "person of color." I have. But there are numerous student groups dedicated to expressing Columbia's diversity. But keep in mind that Columbia and Barnard (to a lesser extent) are HIGHLY selective- you need top grades. And I wouldn't mention your yen to live in Harlem in your application essay, either. Admissions reps don't like stuff like that.
I can't generalize about the people. You need to experience that yourself. In general I believe that people are the same no matter where you live. You'll run into nice ones and you'll run into assholes.
There are tons of interesting places all over Manhattan. I wouldn't just stick to Harlem. In the immediate area you have the Apollo Theater, St. John the Divine, Tom's Restaurant (aka the diner from Seinfeld), Morningside Park, and the Museum of the City of New York.
Everything is expensive in NYC, but yes, McDonald's does have the dollar menu. You can also get a slice of pizza as long as your arm at Koronet Pizzaria for $2.50.

First City College has a campus in Harlem.
Harlem is a section of Manhattan that is in transition. It is predominantly Afro American but the lower part of it has a lot of college kids and young people of all races. It is getting expensive, as is all of Manhattan. My neice who has red hair and blues eyes lived there for several years with no problems.
In Manhattan you have theaters, restaurants, night clubs, concert halls, opera, ballet, extensive shopping, libraries, museums, parks.
You don't need a car, in fact, you are better off without a car. The subway can take you anyplace in the city much quicker than you can get there by car. As far as festivals, just about every ethnic group has a festival, St Patrick's Day, Steuben Day, San Gennaro in the Village, St Anthony in the Bronx, Puerto Rican Day, Dominican Day, Columbia Day etc.
If you have the money, there is something to do 24 hours a day. If you don't have the money, there are many free things such as Shakespeare in the Park, Philharmonic in the Park, the Central Park Zoo, fireworks displays on July 4th.
Despite their reputation, most New Yorkers are friendly.

Why would you focus in on Harlem without a college in mind? If you're thinking of Columbia, which is in Harlem, then that would be a start. But wanting to go to Harlem as a first priority is bogus.
With much of Harlem being comprised of housing projects, is not safe at night, but it is very diverse. I wouldn't call the people friendly.
It has great transportation.
The food is good. Good spanish food. And I can't believe you just asked if there is a McDonalds in Harlem. THERE ARE FOUR MCDONALDS ON EVERY BLOCK.
From the sound of it, you do not want to go to Harlem. I think you just listened to too much Dip Set.
