Gotta take motorcycle gas tank off to mess with cap, what else can/ should I do while it's off?

Flush the gas tank, possibly clean out your carburetors if equipped. Install new fuel lines and inline fuel filters, check/replace your air filter and possibly replace your spark plug wires. Basically give your bike a minor tuneup.

You should be able to leave it in the "ON" position and and remove your tank without the fuel coming out. If you have a "PRIME" position that means your fuel petcock should have a vacuum operated diaphram that does not allow the fuel to flow past the petcock unless the engine is running. There will be a smaller diameter vacuum hose going from one of the carbs to the rear of your petcock, this is where the vacuum comes from. The "PRIME" position on the petcock is used to bypass the vacuum diaphram... say you just had the carbs off and cleaned them, you re-install them and there is now no fuel in the bowls, the bike will not start therefor there will not be enough vacuum to open the diaphram. You turn your selector to "PRIME" and it lets the fuel into the carbs without needing any vacuum, you can then start the bike and once you get it running, set it back to "ON" and be good to go. You can also drain the tank through the petcock fuel line while it's off the bike by using the "PRIME" position.

Make sure the inside of the tank looks good...any rust? Take the opportunity to clean & seal it.

Depending on the model -
Check/change air cleaner
Check/change spark plugs
Adjust valves
Top off the battery with distilled water and charge it.

Check and replace any faulty wires.
Remove all wires, sand and paint the frame.
Replace the wires.
