I dated my best friends brother for a year. The best thing to do is tell her. Best friends should not keep any secrets. Also, another thing, you will have to make sure that you spend the same amount of time with the both of them.

If she doesn't know, then it is not ok. Because, brother and sister have a special bonds with each other. If you kept it a secret from her, she's going to feel that you are using her to get to him. Also, she will lose her trust in her brother and their relationship could be sour. Specially if you and her brother break up, then not only you will lose your best friend, you will lose her trust and him too.
So, be honest to her and tell her that you are serious about going out with your friend's brother and that you will take every precaution not to hurt him or her.
