i already have the flooring installed ,but i am wondering if
i can build the closet on top of the floor or if i have to take it up.

Laminate flooring is a floating type floor. If you build a closet on top of it, it will not float. It needs to be able to expand and contract. All is not lost. Mark out where your bottom plates will be, add a quarter of an inch to each side, and cut it out with a skill saw. Be sure to carefully adjust the depth of the blade as not to cut the vapor barrier. Now you can ram set your plates to the floor, and the trim will cover the gaps. The floor will expand and contract, instead of buckling.

I think you can, because we have a closet with stick on hard wood floors

Yes you can,leave it in place and cover it first with plastic before you build the wall,then place cardboard over the plastic not to ruin the floor.
