The 700 Nitro Express is one of the biggest in terms of bullet diameter. There are some slightly bigger but I think most are wildcat cartridges and not really a mass produced commercial cartridge.
The .50 BMG has the most energy in a commercial load but handloaders can push other cartridges higher than the .50 BMG.
10 gauge shotgun slugs and old 8 guage slugs (for industrial use) are larger still going by bullet diameter but are not as powerful in terms of energy.
If you want to get into military stuff, 30mm (30 x 173mm) and 40mm rounds have larger diameter bullets but are fired in canons and aren't commercially made obviously.

.50 BMG is the most powerful.
10 guage shotgun is the largest commercially purchase-able bore diameter. Larger even than .700 Nitro but the actual projectile(s) are not. 12 gauge is .729"
2, 4, & 8 gauges have been made but are far more rare.

12.7mm (.50 cal) is the largest diameter bullet that is sold commercially. Anything over .50" is considered a "Destructive Device" and requires a special process from the government to have legal ownership.
Although technically the diameter of the 12.7x99mm round is .510", the barrel of the rifle is .50" and when fired the bullet is stretched slightly and conforms to the barrel making it a legal diameter.
Edit: True a .700 nitro is the biggest diameter bullet, but at $100 a bullet it is far from commercial.

Technically, you asked for the largest caliber "bullet"....so a .75cal musketball counts and would be the largest. I've never seen any larger caliber bullet for sale anywhere.

The 50bmg does NOT carry more energy than the .700NE. The 50bmg however does have more fps. The impact of the 50bmg is less than half that of the 700 NE.
Others are made bigger. There are none to be consitered commercial.
