first get zeroday protection for viruses (virus protection based on activity paterns not definitions) cyberhawk good and free
get anti-spyware spybot s&d, ad aware, and windows defender.
if you dont have AV get avast 4 Home best free out there in consumer reports and pc mag(beats AVG and is more user friendly)
If you want the best suite money can go with either symantec NAV 2007 or Zonalarms suite.
next this can be complicated. go Start>run>msconfig
go to the startup tab and disable all the stuff you don't want.
next if you really want to do some tweaking go to the services,
and disable everything you don't need. (if you don't know what it is leave it or google it)
then click apply and the reboot
when you reboot it will tell you that you changed the setting click the hide never show again
click ok and you might have to reboot again.
now you should have a sleek computer running at ~190-215Mb of ram usage on idle. with AV and anti-spyware and all the stuff on your task bar.
other than that just keep your desktop clean of any downloads.
everything on your desktop has to be loaded when you startup.
and use a reg cleaners from the first guy BUT make a system restore before you start as they can screw up your system.(believe me that experience talking)

Many good answers.
Don't forget to defragment your computer on a regular basis, either.
Here are some of the basics: Protect your computer from spyware (e.g., Zone Alarm, AVG, Norton, etc.) via firewall and antivirus protection.
Keep your computer updated with latest Windows updates (patches problems and security risks on your system).
Registry cleaners and CCleaner help clean up unnecessary clutter on your system. (Not the best, but Start>Accessories>System Tools>Disk Cleanup can do some good.)
Defragment - As old documents/programs are deleted and new programs, documents, etc., get saved to your computer, your computer gets fragmented (long boring explanation fo rthis process). This slows down performance and increases the chance for errors.

If your computer is not working properly while you are working on it, it could be a problem with device drivers, hardware or software.
