The builders are the religious doctors of the law. They rejected Christ, and Christ became the 'capstone' (culminating icon) of their religion, i.e. their Messiah.
"Everyone who falls on that stone will be broken to pieces, but he upon whom it falls will be crushed"
Those who deem Christ as Evil will be judged harshly, but not nearly so harshly as those who, by their abominable actions and sins, will be deemed evil by Christ.
Second Interpretation for the second part:
"Everyone who falls on that stone will be broken to pieces, but he upon whom it falls will be crushed"
Those who come to Jesus and confess Him ('fall down before Him') will be persecuted, and heavy temporal sacrifice will be demanded of them. However, those who don't come to Christ will make an even greater eternal sacrifice, one that 'crushes' them forever.

One needs to read the other scriptures, that lead up to that scripture, Jesus was speaking with the chief priests and elders of the temple in Jerusalem, Jesus related to them the illustration of the vineyard owner who lent his vineyard out to cultivators, and when the owner sent his slaves they beat up one and killed another, the third one was stoned, then the owner sent his son, whom they killed, what did the priests and elders believe would happen to the evil cultivators? Why they answered the owner would destroy them, because they were evil, then Jesus says: "Did you never read, in the Scriptures, 'The stone that the builders rejected is the one that has become the chief cornerstone, {who is Jesus} because the chief priests and elders had stopped teaching God's word in the way it was meant to be taught and began teaching the Traditions of Men insted. The kingdom of God would be taken away from them, and would be given to a nation producing God's fruitage, [the way he wanted his word to be taught] Those who had not been faithful would not find refuge with God anymore, and would now be destroyed. Which happened in 70 C.E.

Jesus is the rock the foundation on which I stand.
Jesus is the stone the builders rejected.
The jews wanted someone to deliver them from roman rule. In fact a politician and King.
Jesus was rejected for He came to deliver his people from the snare of the fowler.
Those who reject him are dead or fallen over his Words unto spiritual death. Blinded to the truth. Deafened to the truth. Hearts hardened to the truth.
