"Speak clearly!" when addressing to somebody who said something you didn't understand because he/she was mumbling, etc
"Say it straight!" (or also "Tell me the truth, don't joke with me!") when you want somebody to be frank, sincere to you; for instance when somebody is hinting that you're mate may be dating somebody else but he/she's not saying it straight but just "suggesting" it (for any reason, good or bad) you say "Parla chiaro!"

It's a bit strange to work out! Parla comes from the present indicative parlare (to speak) Parlo _ I speak Parla ( you speak formally) Parli you speak informally. So the word parla is you speak. Chiaro is translated as light coloured. But the present indicative chiarire ( to clarify, make clear, explain.) is chiaro I clarify, Chiari you clarify ( informally), and chiara is you clarify so I believe it is parla chiara which then would be speak clearly.

The sentence sounds a little bit strange in Italian. Is it just a bit?
However I will try to explain it clearly.

If it is "Parla chiaro!" it means "speak clearly/speak up"
If it is "Lui/lei parla chiaro" it means "he/she speaks clearly"
