Ride a bicycle at least 40 miles a week. It does wonders.

Somebody already said it. Find something you enjoy on that level and just get better at it. Explore different things. For me it's yoga, martial arts, biking and walking. Hopefully exercise will become something more than some kind of unpleasantness to be endured for the benefits of fitness, but rather to be enjoyed while in the act of doing it. Fitness becomes almost to be as just an added bonus. Moving freely and pain free with energy through life is a wonderful thing.

ride a bike, everyday if possible
run a few miles, everyday if possible
take a class at a gym (aerobics, spinning, etc.)

many different ways, no "best" way, do what works for you and keeps you motivated (in other words, do what you like and stick to it!)
