I would say, "I really don't appreciate you making such personal comments about me, I don't think this is the time or place. I really thing you are being inappropriate."
Chances are, you won't create bad blood b/c one of two things will happen:
1) your co-workers will realize that they made a fool of themselves for making such a comment
OR
2) they won't ever make a comment again. Chances are they might be testing you to see if you are passive to their bullying behavior. They want to know if you have the ability to stand up for yourself, if need be. Don't let them walk all over you.

From reading your question, I am assuming that you have known these people for some time and perhaps they are unaware of how rude it is to comment on ones personal appearance no matter how long they have worked with you.
If it happens again, I wouldn't be rude back, simply say: "now that is rude" and keep walking/working whatever.
They sense your sensitivity and for some reason keep at you.
You can always lose weight if you choose, others need therapy.

Some people crave drama, others are so desperate to feel above someone Else, both groups like to pick on people to get what they need.
Lot's of things you can do. Ignore it. Inform them politely that it is not professional, or conducive to the team spirit of the workplace.
Or you could make up something outrageous to shut them up, like' "I just finished my chemo therapy, and I am ravenous. My Doctor says not to worry, my appetite will return to normal."
Try to control yourself, you might get in trouble, and I don't think your boss will be sympathetic when he/she asks what started it, and you say "she started it! She called me fat!"
See opening statement about team spirit in the workplace. LOL!
And be careful with the smarty pants answer, someone you really do like might hear you had cancer, and send you a congrats, you are off chemo bouquet. And you will back in front of the boss, explaining why your team member does not speak to you anymore, and insists you owe her $100 for flowers.
Hope I made my point clear. do anything besides ignore them, and you will be the one in trouble.
Good luck! and by the way, according to the division critic, my hair is not to her liking. "Can't I fix it some other way?" Hers is 1950 platinum blond, done upsweep in the do of the era. All she needs is a poodle skirt and bobby socks. Here is the difference in us, I would not think to tell her, or my co-workers what I think of her.

Try being civilized first. Tell them on the spot they've crossed the line. Don't get loud with them or create a scene, why would you jeopardize you're job for someone like this? If it happens again you might have to tell her you can lose weight but she'll always be ugly!

HA! You are getting a bit of the treatment women dish out to men all the time. It is their RIGHT to butt into your business and say whatever it is that they want to say.
You must tolerate this treatment, because "They Are Women".
Or not. I think you are smart enough to tell them off with an audience so that they will think again before doing it to another.
