Amen is a derivative from the Hebrew verb aman which mean"to strengthen" or "Confirm". The word has been used in both the old and new testaments and it is a way of either opening a prayer or closing. As Christians we confirm our faith when we pray or when we make a statement of faith, using the word amen is a way of just strengthening or confirming our faith.

I am Wiccan and I say .. So mote it be.... It is simply an appeal to your Deity, and at the end most of your energy go's into the last word ... In this case Amen..

It's just a word that you use to say "OK God, that's all." So everything you say after that isn't included in the prayer. I don't say it very often anymore because I think the Divine know when I'm done praying.
