Depends on what your personal interests are, because the city has just about everything. You should see at least one Broadway show. Buy discounted tickets at tkts in Times Square day of show. Opera at LincolnCenter if you like it. Many great restaurants--Tavern on the Green is a favorite for tourists and locals. Walk around Greenwich Village and SOHO. Central Park, but there are other parks that are nice and quite safe like Riverside Park (weather permitting.) Take a cruise by the Statue of Liberty. Your hotel will have a copy of Where magazine in your room or lobby so you can book these things. Have fun.

Go to times square, walk around, buy a ticket to a broadway show for your fourth day. Go to Madam Toussaud's (couple blocks away) then walk down fifth avenue towards the empire state building (going into all the interesting stores along the way) then go to the observatory of the ESB. Leave the building and take the subway (you can do it!) to ground zero. Take a tour of the area, go to St Pat's to see the pictures, paintings, etc. Go to fulton street, by the water. Go inside all the stores, the mall, def go see the Bodies exhibit with the plastinated dead people if its still up (you'll see posters, or just ask).

Day two you should go to central park, walk around for a bit, find a nice place to sit and eat. After 3 hours or so head to the museum of natural history (amazing museum) then go to the metro museum of art.

Day 3 you should visit china town, eat some great authentic food, buy and eat lots of fresh fruit as you walk along. Walk towards little italy. Have Lunch.

Take subway to the village.

Day 4, go to the statue of liberty. It should be open by now but if its still closed then take one of the boat rides that go around it. Go back to times square at night and see the play you bought a ticket for.

Times Square, NBC Studio Tour, Central Park, Brooklyn Bridge, a ball game if it is the proper season, the statue of liberty, and be sure to take in a broadway play....

hit all the tourist traps (time square, the statue of liberty, ground zero, etc.) and shop til i drop :)
