The question is :
Que=what
insistio=insist
tu=your
profesor/a=professor
de=of
clase=class
ayer=yesterday
So, I'm thinking the question is something like "what did your professor ask you to do yesterday?"
So, you could answer "Mi professora insistio nosotros estudiamos ayer" which is "my professor instisted we study yesterday" OR....you can say "Nosotros leeramos un libro ayer" which is "we read a book yesterday"
I hope this helps, I'm no expert but I think this is right.....

It seems your teacher was insisting on something.

The question is:

Yesterday in class, what was your teacher insisting about?

so, you answer something like:

she insisted on taking a test.
ella insistió en tomar un examen.
