I heard a story about it: Mona Lisa means "My Lisa". Her real name was Lisa Gherardini. She lost both her husband and her son in a very brief period of time. No one can make her laugh anymore. So Leonardo Da Vinci gathered several comedians to make her happy, she finally smiled, hence Mona Lisa's smile.
Other sources say she did to celebrate the birth of a child. (notice her bulging stomach)

Hard to say seeing as no one really knows for sure who it is. I even heard a rumor it was him in drag, but I'm not sure that's very credible. :)
Portraits back them were commissioned by wealthy people a lot the time. They would wear costly clothing and have their lands painted in the background. A commissioned portrait for a wealthy lady is probably the most likely origin of the painting.

He was probably commissioned to do so by the sitter's husband however, if this is the case, he never fulfilled the commission as he kept the Mona Lisa with him until his death.
