I have poured a concrete slab and plan to build a small shop. The shop will only utilize a portion of the slab.

I am using treated 2x4's. Will this sill sealant or asphalt type of material prevent any water from comming in?

First, you want to use for that a Treated 2x4, and put a 3 1/2 inch wide insulating material under that 2x4

That seals it off and allows expansion and contraction. Purchase that 3. 1/2 wide material at Lowes or Home Depot It is not very expensive and contains an asphalt material or fiberglass insulatin and is about 1/4 inch thick uncompressed.

No glue caulk is ever used.

Typically, one would use a sill sealer, which is a 3 1/2" wide closed-cell foam made just for this purpose. It is sold at building supply houses.

I reckon caulk would be better than nothing.

I presume, by mud board you are speaking of a "sole plate" which is the bottom board of a wood frame wall.
If I am correct, then the answer is yes, but a better substance to use would be construction adhesive, like heavy duty Liquid nails.

You should use sill seal and a good silicone caulk (lots of caulk)
