You need to use an administrator account to edit Windows registry. In addition, you need to disable any security program which prevents any edits to the registry. Usually, antispyware programs lock the registry (e.g. Webroot's SpySweeper, Spyware Doctor, Spybot...etc.). Also, host intrusion protection software lock the registry (e.g. Winpatrol, Cyberhawk, Arovax..etc.) I don't know what security software you use; so, i can't tell you where to find the controls.

The registry being locked can be a policy enforced on your computer or a worm or a virus infecting the computer. However, to open the registry again, follow the following steps
Click Start>>Run
type in gpedit.msc
Expand "User Configuration". Select "Administrative Templates" and select "System"
Under system, there is an entry that is labeled, "Disable registry editing tools". Double click this entry. Select Enable. Hit Apply. Select "Not Configured" and hit OK. You should now be able to open regedit.
