No. All that needs to happen is for her to have a penis in her vagina. Some girls don't have a hymen, or it was broken in an accident or during sports.

The medical definition of a virgin is someone who has never had sex although it can mean different things to different countries and cultures.

The hymen (the thin layer of skin around the vaginal opening) can be broken such as thru a fall, or as in my case when I got my ver first pelvic exam and pap smear when I was 18 because the dr used the regular sized instrument called a speculum for that exam.
