Not sure exactly, but can give you an idea. On average, the FAA controls between 51 - 53,000 flights a day over the continenal US. If the average daily total is 52,000, that's 2,166 per hour over a 24-hour period. If each plane carries 100 passengers (a very rough guess) that makes 216,600 people in the air at any one time. Those numbers relate only to those aircraft on instrument flight plans, controlled by the FAA. Add flight crew numbers and general aviation aircraft on visual (uncontrolled) flight plans and it might be a bit more.
