Is there a huge advantage to having 5volt over 2volt preamp output voltages?

Depends if the unit is CEA-2006 compliant. If both systems are compliant, then there is a slight difference.

The lower the voltage, the more headroom is available. However, the higher the voltage, the less the amplifier must amplify.
Usually, the lower voltage is cleaner at higher volume levels (less THD), but marginal.

Although the audible difference will go unnoticed as long as the gain of the amp is set correctly in either case.
