If talking to someone you know well:
"Do you understand?" is "¿Entiendes?
If talking to a stranger: "¿Entiende usted?" is better
I understand is "Entiendo"
I don't understand is "No entiendo"
In some films, eg cowboy films, you may have heard people say "Comprende?". This word does also mean "understand" in Spanish but "entender" is more generally used in the sense of to understand a language.

You can use 2 verbs for it Comprender or entender both mean the same to understand so.
I understand would be = Entiendo.
Comprendo
If you are referring to a subject or and issue you would say :
Lo entiendo/ lo comprendo. Is like saying i understand it.
I dont understand would be = No entiendo
No comprendo.
Or as i pointed before No lo entiendo/ no lo comprendo = i dont understand it.
Depends who is the speaker from would rather use comprender or entender, also depending on the situation. In Spain we use both.

¿Entiendes? = Do you understand?
Yo entiendo = I understand
No entiendo = I don´t understand
You must understand(entender) something in order to be able to comprehend. (comprender).
But in principle you can both any of them. It is more or less the same.

i'm living in spain and most people would say "entiendo" for I understand and "entiende(s)" for do you understand? The verb comprender isn't used that much.
