Autism usually needs to be diagnosed by several specialists, there are many different pieces of it, as well as the fact that it usually presents with another disorder. (40% of the time, there's something else going on besides just autism)
The best general definition of it is that the person is trapped inside themseles. You'll see children that don't make eye contact, don't empathisize with others, and can't communicate clearly.
Higher functioning autistics (aspergers) will usually present with extereme sensitivities-- light, sound, and smells can upset them and cause massive headaches, even if they're just barely noticable to you.

If it is a child you are talking about, you can take them to a neurologist who specializes in the testing of children with problems.
You can also check out the website www.autism.org, you can find all the check list there (they are quite lengthy) There is several different things that have to line up in order for it to be called autism.

That has to be medically determined, by a doctor. They usually catch it early in children and ask about it if they have behavioral problems and attention issues. Try asking your doc, they will answer the questions you have better than I .
