Depends on what type of "handgun" you are looking for.
You can get a break open or bolt action single shot pistol in varying calibers up to .700 Nitro Express. I have never heard of one chambered in .50 BMG but I'm sure its possible.
.50 BMG has higher velocity, but .700 has more bullet weight and energy.
These guns are very expensive (1 round alone can be $100) and I can barely imagine the recoil.

For revolvers the S&W .500 is currently the largest production chambering available.

.50AE is the largest semi-auto caliber.

The Maadi Griffin is a maker of single shot .50 BMG rifles and carbines not a handgun. Although the Model 92 carbine model is unbelievably short.

You are correct the Maadi Griffin did shoot a 50 BMG round the problem was the ATF didn't like it so they arrested him and threw him in jail he's out. But unless your extremely lucky you might not find the plans Because there was no paperwork involved with the selling of the plans. the ATF is constantly looking for people who either have or who have made this gun. If you want the largest round for a handgun S&W makes a .500 hunting round that'll give you a wakeup call when you shoot it.

Nobody makes a .50 BMG hand cannon. There is a .50 Beowolf for use in rifles that is bigger than Smith's .500. Some specialty handgun makers can take your Ruger and modify it to shoot some might big rifle bullets, the .475 Linebaugh (I think it's called) being one example, But they are expensive to get and recoil is like getting hit with a sledgehammer. Not practical for casual plinking or target shooting. As for the gigantic .50 BMG round, there are several specialty makers of single shot or bolt action rifles and a couple of semi-auto repeaters out there, They aren't for the weak of body or the average hunter, though they can be used for very long range hunting up to 1 mile with scopes.
Then there a few other new rounds like Smith's .460 high velocity and Ruger's .480 Ruger that shoot close to, if not better and faster than, the .500 Smith & Wessons. Big money can get almost anything you can imagine. Thompson Center makes a grip frame you can swap barrels on to change calibers up to high powered rifle rounds. They're expensive and mean't for handgun hunting. Look in shotgunnews.com for ads about hand cannons and special order boomers.

Currently, the largest handgun or revolver caliber I'm aware of is the .50.

Fired the S&W 500 this past summer, the recoil was BRUTAL as was the Desert Eagle.

Doubtful I'd ever personally want to buy either of them. The ammo is too expensive for target shooting and I don't see myself doing much pistol hunting......

Yep, the guy above me put it right.

The most powerful commercially produced handgun cartridge is the .50 S&W.

.50AE is the most powerful commercially produced round for semi-auto handguns.

There are niche rounds like he said such as .700 nitro express, but yeah hehe the guns to fire them cost like 75000 British pounds.

Powerful indeed, but with rounds like that I think it would be a waste since I'd never get out to shoot it, far too expensive.

There's a video on YouTube of a guy shooting a 600 NE revolver. Nobody would shoot but one round from the thing, so I don't know why they'd make it in revolver form, but I guess somebody always has to push things, whether it's sensible or not.

.50AE may be the largest round in an auto-loading pistol, but not the largest in a pistol. T/C contender makes a pistol with inter-changeable barrels that go up to 45-70.
