me and my friend are building a go-kart and if the hand brakes (peices of wood that will slow down the wheel) screw up and we cants stop what shall we do

An emergency cut-off can be rigged with a simple (normally open) momentary push button. Connect one side of the push button to the spark plug with 12ga wire. Conect the other end of the push button to the metal frame of the engine with 12 ga wire. When you push the button, the spark will short to the frame and the engine will die.

Most lawnmowers have a seat switch that cuts the engine,This can be moved to the starring wheel where it can be accessed.If you don't have this hock a wire from the spark plug or oil alert to the engine through a push button switch,this will stop the engine.Be sour test this before going any where . If the drive line wont stop or brakes wont stop you ,If chain drive a wood brake over the axle sprocket will work,A solid rubber mat that drops in fount of the back wheel succoured to the frame will stop you to. Always test new to you ideas before really needing it and at low speeds with safety gear.

Either add a second brake system of the same type (though pieces of wood don't sound great, for a budget go-kart coating the wood with a strong rubber might work better), or a floor brake at the rear (would need to be very strong and fixed securely to the chassis), again a rubber material would work.

Could do it with a simple lever mechanism (sprung so it stays in place) and a release handle somewhere close to the main brake?

Use explosive bolts to attach the wheels. When you need to stop, hit the bolt ignition and the wheels will fall off. If that isn't an emergency stop, then I don't know what is.
