How we can place a PASSWORD on a sub-directory? Like we do on Excel & word files.?

We creates Sub Directories under My Document that carries multi-types of files. usually on files like wordfile, excelfile etc we can place password to open. So likewise, is there a software or way to provide security to a sub-Directory?

If you're running Windows 2000 or later then you can set permissions on individual folders without needing any extra software. There's an in-depth article about setting it up and managing it here:
