Apart from people who have absolutely nothing but mockery to contribute throwing their oar in? I would say the apostasy that the church fathers warned would precede the rapture and tribulation period.
edit: As we draw closer to the unveiling of the coming deceiver, the stage for his arrival is being set. Jesus, Paul, John and Jude give very strong warnings about the religious climate that will precede the second coming. The relativistic theology being embraced by some orthodox denominations (gay bishops, etc.) and the rapidly growing self centered (as opposed to Christ centered) movements swarming across the planet seem to show that those days are here. Pastors are enjoined from instructing their congregations on many issues facing us today on peril of losing their tax exempt status. And they capitulate. Listen to some of the leaders of the largest churches and count how many times they preach repentance from sin and dead works. I fear so many of them fit the description of those who "have been robbed of the truth and who think that godliness is a means to financial gain." (1st Timothy 6:5) This is the Great Apostasy, and it is the greatest challenge facing Christianity today.

It all depends upon who you ask. Some might say that the biggest challenge Christianity faces is the teaching from a young age that mankind is a huge cosmic accident and that there is no God except the "god" of random chance.
Others might say that the biggest challenge facing Christianity has remained the same throughout the centuries. False doctrine and false religions where the evil one sows tares in the Kingdom and lures people into false religious systems.
2 Timothy 4:1 I charge thee therefore before God, and the Lord Jesus Christ, who shall judge the quick and the dead at his appearing and his kingdom; 2 Preach the word; be instant in season, out of season; reprove, rebuke, exhort with all longsuffering and doctrine.
3 For the time will come when they will not endure sound doctrine; but after their own lusts shall they heap to themselves teachers, having itching ears; 4 And they shall turn away their ears from the truth, and shall be turned unto fables.

The lack of integrity contained within the scriptures, and the lack of a cohesive philosophy by which people can discern exactly what Christianity is. There are so many different denominations of Christians, it is apparent that you cannot agree amongst yourselves what the truth is. If that truth cannot be discerned, how can people be led to God? It is the blind leading the blind. No good can come of it. No spiritual enlightenment, no love of God, nothing but hypocrisy on a wide scale.

Staying true to the message.
I find that there are way more Christians with an unfocused, errant message than there are with the right message. I'll tell you what the right message is, and you compare it to the message of Fundamentalists and others that you read here and elsewhere.
The right message is that no matter who you are, what you have done, you can recieve God's forgiveness through claiming the covering of Jesus Christ. Continuing in the trust of God, you live a life of faith with God's influence upon you and you change according to his will, whatever that may be.

Living a true Christian life personally and publicly. Representing your Lord well so you can following his last command to make disciples. Being genuine. The bible says you will know them by their fruits. I think too often the modern Christian (in general) is leaving a bad taste in the mouth of a "lost" world. I don't mean this to be negative so don't take that way.

oh slow in believing all that the prophets have spoken. It is taking God at His word and DOING it. But in order to do it, we must believe Him. He said we are a kingdom of priests, yet throughout all of the bible, if you were a king, you couldn't be a priest and vice versa, yet Jesus is our high priest after the order of Melchizedek. Melchizedek was both a king and a priest. So if Jesus is a king and a priest forever, and we are called to be like Jesus, the firstborn among many brethren, then we too are to be kings and priests.
And yes, ALL of Gods gifts are for today.

The post-modernistic society that we live in that tells us that there is no absolute truth...its all about what works for you as an individual.
This stray from the belief that there is one absolute truth that is found in Jesus Christ and the Holy Spirit is killing the Christian movement because nobody wants to hear about "giving their life away to Jesus".
In order to overcome, we as Christians need to change our focus and evangelism style to better suit society today.
*a note to skeptics*..............
by "change our focus and evangelism style" I do not mean change our beliefs or change the truth...I only mean tell it in a way that doesn't sound so repulsive.

If God is for us, who can be against us?
The God, The creator is there, watching us and guiding us. So as we depend on Him, we can face anything.
Even though the evil one and His spirits bring temptation, we can do all things thru Christ.
We need our daily bread from our God and prayer to God, we can face any obstacles in our path.
