LaGuardia is the easiest to get to of the three, but it depends where you flying from. International and long-haul domestic flights (Los Angeles, Seattle, San Francisco, etc..) only fly into Newark and JFK.
Air Canada flies primarily into LGA. They fly to Vancouver and Calgary out of JFK. From Newark, they fly to Toronto and Montreal.

La Guardia is the closest from NYC. You take the Grand Central Park Way and you are there. JFK is a bit further..about 30 minutes further without traffic. There is usually a lot of traffic though on the Van Wyk going to JFK. I recommend you go La Guardia.

If you are entering New York City via international flights, I would recommend JFK since the options on getting into the city are much easier from there than Newark, although traffic wise, Newark is somewhat easier in terms of getting into Manhattan. However, if your flight is domestic (or certain Canadian flights), then LaGuardia may be your better bet. It is much closer to Manhattan. The only downside is limited public transportation options since there isn't an easy access to get to the subway from there aside from taking a bus, as opposed to JFK with AirTrain.

JFK has an extra transportation option: Airtrain connects with the NYC subway. Maybe extra lugging all that luggage and a crappy train car, but waaayyy cheaper. $7 will get u into Manhattan.

They're about equal distance from Manhattan. I believe Kennedy is in Queens and LaGuardia is in Jamaica NY. Flip a coin

LaGuardia is closer to Manhattan, but you'll have more flight options out of JFK. (And, LaGuardia's runways are REALLY close to water. I don't fly out of there anymore if I can avoid it.)

La Guardia is easiest to get to. (Newark airport -- EWR -- is better yet.)

check with your airline, the closest stop may be not where you`d want to be if push came to shove, convienience is what you are looking for. taxis can be quite espensive to rent if you dont do your homework beforehand.
