I recommend starting in Times Square. Fifth Avenue, FAO Schwartz, Rockefeller Center, Broadway, and many other attractions are all about 4 blocks from Times Square. Later, you should travel to Southside Seaport. This port has had many movies filmed in and around it. Also, a ferry goes from this port to the Statue of Liberty. Even though the price of the ferry is steep, I can guarantee you that as you pass Ellis Island, you will feel like the early immigrants did. If you do have a chance, i would suggest going to a broadway. Also, If you get a chance, eat at SPQR Italian restuarant in Little Italy.

Take the Circle Line cruise around Manhattan - you get to see a lot, including the Statue of Liberty, the bridges, one of the stadiums, Brooklyn etc in a few hours. But take a jacket as it gets chilly.
Wander through the lower part of Central Park.
Go up the Empire State Building for a view of Manhattan from a different perspective (it's open until late).
Head down to Times Square after it gets dark just to see the lights. There are some great restaurants in the nearby area too(45-47th Streets around 8th/9th/10th Avenues).

dodge long strains so stay far off from the Empire State development and the Statue of Liberty. bypass to Canal street in Chinatown. Then to Little Italy on Mulberry street and start up heading north to Soho. you notice a lot of interesting paintings galleries and shops there to maintain you busy for hours. And in case you decide on you should maintain strolling north to Washington sq. Park in Greenwich Village.

A one day stop in NY! Well to get a sense of it. I'd recommend a Grey Circle cruise tour around Manhattan, followed by a Broadway show in the evening before or after having a bagel & lox at a NY deli.
