You will have to apply for a new replacement title,

Not a big deal. just go to the DMV and tell them you need a replacement.

Bring the vin# and your ID,

If your father titled and registered the vehicle in his name, then you don't need to contact the previous owner for anything. Your father can just get a duplicate title from the DMV. But is sounds like your father didn't put the vehicle in his name and that is why you have to get a hold of the lady who sold him the car. And that could be a problem if you can't get hold of her. Contact your states DMV and see what they suggest. Different states have different rules and regulations about getting a duplicate title.

the jurisdiction issuing the title keeps a record and you can always get a duplicate (for a fee). you can take your insurance card or your registration card. you can even physically remove the tags from the car and bring them in. they will run the numbers and that will bring up your registration. with proof of ID, they will give you a duplicate title.

note: a replacement title is used if the original title is somehow spoiled (wrong mileage, wrong color, wrong VIN, etc) if everything is correct, you want a duplicate-not a replacement.

You just need to show proof of ownership and you should be able to request a duplicate title. I would call down there and see what they require as proof.
