The best workout routines combine cardio and weight lifting. Women usually do more cardio and less weight lifting because they don't want to bulk up. Men usually do the opposite because they want the muscle mass.

You'll know it's working by waiting it out for a few weeks, and looking at your results. Being sore after a workout is normal if you're just starting out, or if you recently increased your routine, or tried a different excercise all together that worked a different muscle group. If your workout leads to more muscle definition or some weight loss, whether a small or large amount, then it's working. Also, if you're not dieting or trying to lose weight, you'll know if it's working by just feeling better and healthier.

I know that my workouts are working when my clothes start to kinda hang on me. I rarely look at the scale - muscle is denser than fat and that can be misleading.
To actually answer your question, if your muscles are sore after a workout, it's not always a bad thing! As long as they aren't so sore that you can't get out of bed, you should be okay. You've totally overworked them if it hurts to raise your arms so you can style your hair or if it hurts your chest and stomach when you laugh. (been there, done that!)
When you do feel a little achy, that means that your muscles are "reinventing" themselves, so to speak. They are changing and becoming leaner (or thicker...depending on your choice of workout). At that point, it's important to take a day off from exercising. That allows your muscles to completely recover and finishing changing. Resting helps you see your results faster.
