There is not one that will always be cheapest. You have to shop around. Seasonal demand, length of stay, and fare sales can affect prices greatly. Also, depending where you are in the world, Priceline, Hotwire, et. al. might not even work, because they are set up for business mostly with North American passengers. As far as cheapest airline tickets, probably the airline's website directly is the best option. I fly a lot for work so I book a lot of travel. I always check expedia.com or orbitz.com or one of those sites, and see which airline has the cheapest flights. Then, I go to that airline's website and pick the exact same flights, and more than 99% of the time I get the exact same fare. Except, most of those websites charge a "booking fee" of $5 or more so it's actually cheaper through the airline. And many airlines guarantee that you will get the cheapest fare through their own website. I know American Airlines does this and probably most others do also.
Also, many of the "low-fare" airlines (Southwest, JetBlue) do not list their fares on those websites so you actually have to go to the airline's websites. However, most of the time I find those airlines to be at least as expensive as other airlines.

Cheapest flights are not available online. Travel AGent have more access to rates and packages to make your trip cheaper AND easier.
Not only that, using a travel agent is FREE. YOu do not pay for the travel agent to set up your itinerary - they get their money from the Airlines and Hotels.

use those compilation sites first...like expedia....find out which airline is cheapest and then go straight to the airlines website....i do that and usually get my tickets about $5-25 cheaper
