that depends.... the space shuttle physically going up doesn't, but the smoke and smog that comes from it might work torwards it.( although it's a debate)

NASA says it has measured the ozone above the launch site right after a launch and there is no hole. However, the solid rocket booster exhaust does contain ozone destroying chemicals.

no. its like a submarine through water as the atmosphere is fluid, and the hole created closes up immediately behind it.
