Just tell them it is getting to be too much of a handful with both kids, so you won't be able to sit any longer.

First of all, that's a rediculous amount of time to have to watch someone's kid unless you run a day care. If he's not compensating you, then I would say you have every right to just tell him to forget it. Even if he is, just tell him, that the kids don't get along and that you think it would be better if he found a day care or someone else to watch her.
How old are these kids that they don't go to school? If they are that young, then it is even more rediculous that he expects you to watch her for that amount of time.
Get out of that. You're not a bad person for not wanting to do that anymore. I don't blame you. Suggest a day care to him or something.

Just tell him that your schedule is too loaded or that you are not getting enough time with your daughter... Or that you have some unknow very contagious illness! Lol. Kidding.
Seriously, I had to deal with this recently. I'm a nanny and a mother down the street plotted with the mother I nanny for to babysit for her. I did it once and the boy was horrible. Just said that I don't have enough time to focus on the three I already take care of and that was that.

I would just be honest and tell the father that you don't have the ability to watch her anymore. Or maybe that you would like the freedom to leave the house with just your son. You don't really need to get into the reason that your son doesn't like her. I hope he is paying you for the childcare. Good luck.

That's just it. You tell him you can't watch her anymore...it's too difficult.
You could have some suggestions lined up for places he could take her which would make it easier on you & it would look like you were doing him a favour at the same time. There must be other stay at home mothers doing day care in your area - ask around & then give him the names.

I don't see why you have to give him any explanation as to your son's feelings regarding his daughter. Just tell him that you need a break, and you won't be watching his daughter any more. Whatever he says, do not let his "guilt" you into changing your mind. You can do this pleasantly. Don't feel bad about it.
Good Luck!

Tell him that the Television has been talking to you lately while keeping a glazed stare in your eyes but look past him as you speak to him.

just say that it is not a good day. After twice, they will catch on, no discussion needed.
