tough question. I dont think you can narrow it down to just one question. .so here are the biggies in my opinion:

1. is this all that I am?
2. will i be remembered?
3. will I die alone? Do I want to die alone?
4. what will i be remembered for?
5. will my death be of a violent nature?
6. will the memory of me be honored by my children?
7 when I die will my significant other know that he or she was loved?
Will the Lord take me?
9.how many people will show up for my funeral
10. how will other's lives be affected upon my demise?

this is one of the toughest questions I have ever answered

If this question for yours is answered then you need not ask for more.
"Am I really content?"
Here is the answer:
13 Jesus answered, "Those who drink this water will get thirsty again, 14 but those who drink the water that I will give them will never be thirsty again. The water that I will give them will become in them a spring which will provide them with life-giving water and give them eternal life."

Do read that chapter John 4, to get the whole story and the point made. I really think, we can only be satisfied when we find the truth and have a hope that God gives.

Actually that question is very important, Whether you share it or not is strictly up to each individual but I do feel in sharing ones question(s) can be helpful to others therefore a help in finding answers yourself. To me the most important question, is "What is my life's purpose?" Followed by am I living my life's purpose?

well.. for me, the most important thing to ask is: what is the question, after all? is it my question, the question that decides between life and death, coz.. all other questions are already asked and answered by some body and has not made any difference to me! let us not split life into material, religious, philosophical or spiritual.. spirituality is, being one in thought, word and deed and any thing that upsets this equation is against spirituality and life..

Are we serving God in a way that he approves?
We were created to honor and worship Jehovah. Most of mankind just doesn't care about God. They continue in their everyday life never praying or thanking God for anything.
We all need to take in accurate knowledge of God and his son Jesus to get saved and enter the kingdom of God and live forever.

I can't rank my questions as most important to least.

1) What am I doing right/wrong in life and could I do more/better?

that one is most important to me if I had to pick one.

2) Do my beliefs make sense to me or do I believe them because it is what was placed in front of me.

this one is more something i'd like to ask others I know.

fun question though, made me think.
