Can be hard on your lower back if not properly fitted. Great for Leg strength and cardio. Just take it easy at first, don't run as you normally do.....take it slowly at first

The positives are that you will probably, in the long run, lose more weight and if you get shot your chances of survival are probably a little bit better. The negatives are that you will run awkwardly with it at first and will be putting most of the stress on only a few muscles, you will look like a geek, people will think you are a psycho that is way too in to yourself, you will have officially reached the point where you have forgotten what running is all about.

Positive: Burn more calories, run faster when the vest comes off, gain muscles.
Negative: You will have a very good chance of getting stress fractures, which are extremely painful and very frustrating

Burn more calories quicker. positive
Bulk up in areas that aren't necessary and end up having to carry extra muscle weight while running long distance, which will slow you down and unnecessarily fatigue you. Negative
Also possibility of chaffing under the belt/ vest, big negative.

Positives: increased weight will command more blood flow and oxygen to the muscles to handle the load.
Negatives: If the person is not in excellent physical condition, the extra weight may inflict severe muscle or bone damage.

Postitive - Burn more calories, build large muscle groups faster, build bone strength faster, training muscles to handle more than necessary so come race time they will be less fatigued.
Negatives - Chaffing, lower back stress, STRESS ON ALL OF YOUR LOWER JOINTS.
The biggest negative of running is long term damage to the lower joints, why compound it.

It increases your workout results.
If it doesn't fit right your carrying
all the weight on your shoulders.
Also once your body gets use to
a set weight, it doesn't work as well.
Change the amount of weight in
your vest, from time to time.

I actually have not heard anything good about them, and weights on your ankles I heard that they are bad for you. Just in case I wouldn't try them.
