I like foxfire's idea of a garage sale.
There is another thing we do at our school. I forgot what they called it, but they had a big jar outside of the office at school, and kids would bring their pennies and put them inside. The adults passing by the office had a tendency to slip in some bills.
Or you could have a contest to see which class could bring in the most quarters, and the winning class gets a pizza party.
Another popular thing is to ask local businesses if they would put a "donation jar" on their counter. You provide the jar, with a label saying what the donations are for.

Is St. Judes a hospital? Start a walk, perhaps on Mother's Day. See if you can get a big organization to sponsor you. It is for kids after all so people are likely to give. You need to get an organization committee going. Get your local newspaper out as well. Get a website, start with the schools in your area. It may be small the first year but will grow.
If there is a child you can name the walk after, do so. Get your local politicians out there too. You can do all kinds of things if you think big!
A bbq or dinner dance always works nice too.

We used to have penny wars at my school to raise money. Have clear jars/boxes for each grade or class. If a student/teacher puts a penny in the box, it earns one point for the class. If someone puts a silver coin or dollar in the box, it subtracts points (ie. nickle 5 points, dime 10 points, ect) At the end of 1 or 2 weeks, the class with the most points gets some sort of recognition. We just did announcements, no prizes. All the money goes towards St Jude. It used to raise a good amount of money. Keep announcing the totals over the week to get students involved.

This is a variant of the penny collection.
In our town one school's students ran a penny jar for passing drop-off and pick-up parents. You need a big banner so that people know at once what you are asking for and why.

Ask your local stores to take up donations for you, when people check out they can ask then if they would like to donate and make fliers all around the busiest stores and restaurants.
