In heaven we are the bride of Christ. We are all Christ's and not given in marriage like we were on the earth. The Bible also talks about a day of weeping and mourning on the day that we see our loved ones go to the Bad place.

The Bible tells us in Matthew 22:30, For in the resurrection they neither marry, nor are given in marriage, but are as the angels of God in heaven. In heaven our time will not be spent worrying about marriage, dating, etc. it will be spent worshipping God. He will be our focus, unlike the things of this present earth and present day.
As for your second question, that's tough to say. Some questions will not/can not be answered in this life. I would have to think that yes, it would be sad to realize that you missed the opportunity to help them come to Christ while they/you were alive. But when the New Heaven and the New Earth are created, I believe that our "thoughts and memories" of the old will have passes as well, otherwise the saying couldn't be true that there will be no tears, no pain, no saddness, etc.
That is why it is SO important that we do all we can in this life to lead others to the knowledge of Jesus Christ. There is no better time than the present to do this. We are not guarenteed another day, or even another hour, so if we know of people who are not saved, we need to begin praying for that person daily and speaking with them so that they can come to the truth and not face an eternity seperated from God.

Jesus told the people when asked about the widow married 7 times, in Heaven we are neither married nor given in marriage. Since there are no tears in Heaven, as far as the loved ones goes, I guess you will realize they had a choice to make and made the wrong one.

I believe that family is eternal and that our relationships are meant to last forever. I can't imagine the thought of my parents/spouse/children dying and never seeing them again. I feel so bad for people who do believe this, particularly when they lose a loved one through death. What a sad, sad thought. At my grandmother's funeral, we grieved and cried, but thought to the day when we will be reunited with her. Furthermore, I have seen her since her death on many occasions, particularly when I'm sad or stressed. How can these relationships not be eternal...families are forever.

Heaven is a bribe so you can expect the heaven you experience to be exactly what you want. The people you want to see there will be there in your experience. The only way you might suspect they were not really the people you know on earth is they would be almost perfect. As we know no one we know here is perfect so you might suspect they were a gift to you and not really the soul of the person you knew. However after 1000 years it won't make any difference.

between the time that God returns to this earth and the time when He makes it anew, there will be a time when we will notice that some of our friends or loved ones are not there and yes that will be a sad moment...but then we will be shown why they are not there. then we will understand how God was being merciful in not taking them to heaven. and in realizing they are not there because they chose not to be.... we will cease to grieve for them.

In my humble opinion, those that don't make it to heaven will not be remembered. That's why it is in every Christians heart that they want to see all their loved ones (family and friends) in the kingdom of God. Sadly though, I'm sure there will be some of our loved ones not make it. They have denounced Christ and anyone that does that will not be there.

everything in the after-life is significant in a really obvious way to expanding your understanding. if you saw your loved one in "hell", you'd see that they weren't really there but thinking they were. the laws of physics there allow you to create with instantaneous effect, allowing you to get far by ineffecient guess n test methods, as a month in the after-life is like a second in earth time.

To live a life of no suffering would require that all memory of suffering be erased. Who you were would have to be altered drastically in order for you to never suffer again. The person you know yourself to be right now could not exist in heaven. And what good does it do you to live for an eternity in harmony and peace if you can't remember your life experiences, the beauty of diversity, or the exquisite pleasures of the full spectrum of human experiences because you have lost awareness of who you were as an individual?
