I have a one story extension on the side of my house which has foundations for 2 storeys. I would like to put a bedroom on top with an ensuite - nothing flash. The size is approx 20 foot by 12 foot.

Big factor is the labor cost. Try to ask some local contractor in your area or neighbors that experince these kind of works.

Depends where you live?
Anywhere in counties with easy access & big garden for materials etc not so bad say approx. £35k all in
but if in London you could prob add another 5k to that

I agree with the 30-50,000. On the east coast it is about $180/sq. ft. for an extension.
