You can lock your carry-on suitcases... you'll have to open them for security. Your checked luggage should be locked with TSA approved locks or not at all. If checked luggage is locked and the TSA wants to inspect them, they will break locks that aren't approved.

You can lock your suitcase at the airport or before you go if customs want's to open it they do so with a pen in the zipper anyway, locking your suitcase doesn't protect your stuff but, yes you can lock it at the airport.

After your suitcase goes through security screening you may lock the suitcase, Last time I travled we went through security screening and that is when I locked mine.
