I may be cheating a bit here, but John Huston did two two-character movies 25 years apart. The African Queen does have some Africans at the beginning (as well as Robert Morley, who never left England), and some Germans here and there, but it's mostly Kate Hepburne and Bogart. Likewise, there are a few Japanese, and at last some US Marines, in Heaven Knows, Mr Allison, but the picture is Mitchum and Kerr. It also seems that Mitchum did a two-character play with Shirley Maclaine, called Two for the Seesaw.

Well I believe there have been a few movies that have had VERY small casts... unfortunately while My Dinner With Andre only featured 2 actors dining, there were also a Waiter and Bartender (so cast of 4)...

For a cast of 2 I recall Alan Rickman and Madeline Stowe in "Closet Land"

Both excellent (and very different) films by the way...

I'm sure there must have been others...

There is a movie called Night, Mother with Sissy Spacek and another woman that plays her mom in the movie. It's just those 2 in the movie. Sissy Spacek tells her mom in the beginning that she has decided that she is going to kill herself and her mom spends the whole movie trying to talk her out of it. There are like seven people int the credits but like 97 percent of the movie is just those 2 talking. Here's the link.
