John Wayne has been in 142 films, however....
Mel Blanc, the voices of the infamous Bugs Bunny and Porky Pig has been in 809 films with an extra 81 voice parts in films.
If you're looking worldwide though, Indian actor, Adoor Bhasi, has been in 549 films.
So American and European movies, the (i guess you could call it 3-D) record goes to John Wayne
For voices of cartoons, Mel Blanc
And worldwide actors, Adoor Bhasi takes the cake

As far as actually appearing in the film (not just a voice, etc), I believe it's John Wayne. He's credited with 142 movies. Now that's really True Grit. :)
