If a man asked me: are you a student?

How do I answer him?

No, I'v been working
No, I'm in work
No, I'm a worker
which is correct? or you have better suggestion?

"No, I work" is a reply I often come across. Alternatives are: "No, I have a job", or "I work for...." (the name of your employer or "I work as...." (giving details of the kind of work you do).

When you say "I'm in work" it's always as an alternative to being unemployed, so this is probably not the right impression to convey! To say: "I am a worker" gives the impression that you are someone who works hard, as opposed to others around you who don't! People don't tend to describe themselves as workers except in that sense and in certain political contexts.

There's nothing wrong with "No, I'm a worker" but a better answer might be "I work with (name of company)."

"I'm a worker" would be correct if you are a worker and not a student. "I'm in work" would never be correct.
