Given that you will need transportation time and time to go through check-in/security, the best idea would be to pick one specific place/attraction that you want to see and not try and do too many things. Or, try and stay in one location - i.e. the Times Square area where there is a lot to see and do instead of hopping all over town.

You will be suprised at how fast the time goes by, and how even just walking around and waiting on line for certain things takes much longer that you planned.

Try to get tickets to "timed entry" attractions, like "Top of the Rock" or the Statue of Liberty so you know that your waits will be minimized.

Also, keep in mind that if you have to travel during rush hour times (7:30 - 9:30AM and 4:00 - 7:00PM) there can be very heavy traffic between the airport and Manhattan, so leave yourself extra time. You need a MINIMUM of one hour at most times of the day.

Lastly, there are no storage lockers of any kind at JFK to leave your luggage for the layover!! Make sure you can check your luggage through before you leave the airport because you probably won't want to lug it around all day.

I know this isnt a specific answer but in 10 hours you can see anything you desire, do some research. Since I've been to Greece myself (have fun on your 10 hour flight btw) I'll assume that you'll be flying out of JFK which is pretty close to the heart of the city. Times Square, along with the rest of the island of Manhattan is well within your grasp in a 10 hour time frame. The only issue is you should probably pick one section of the city to visit and stick to. But really, why would you go anywhere than Manhattan anyway?
