florence of course is a must.

if you can rent a car and drive from florence to siena, the land scape is wonderfull, and every little town is beautiful (san Gimignano,montepulciano...u name it). food is great (in florence go to Il sasso di dante, right next to the duomo.dante used to seat on the stone where now the restaurant is built on it. In siena i don't remember any specific restaurant, but order PICI,local fresh pasta).

whatever city you decide to visit in Tuscany you will be pleased
just make sure you go to florence and than you build your trip from there.

Florence offers a bunch of tours. I am from florence and i took the biking tour in the chiati.i was told i was the only italian who ever did it. i was amazed, it was such a great experience.therefore i assume for a foreign it must be a wonderfull thing to do (good view, food and wine)

if you go in the summer time than lucca will be great, you can enjoy the city and take a 15min train ride and be at the beach (viareggio is the most famous location around there. lots of clubs as well if you feel like partying)

have fun

Tuscany is beautiful. I lived there for almost four years just West of Pisa. Lucca is great shopping and food. Their baked goods are to die for! Frienze is the must go to city. I have visited 36 foreign countries and Frienze is the best city in any of them by far. Pisa and Via Reggio are also nice. Pistoia has an interesting zoo. Pollen is bad in April and May. June through September is tourist season. Weather and prices are best in the October-November time frame. Nude beaches in the Summertime so if you are taking kids be ready for a lot of questions.

Tuscany is beautiful with great food. Lucca is a nice village that has been named in a lot of tourist mags as a place to see. I'd also just wander the backroads of Tuscany...

If you go to Italy, Tuscany is "must".
Best towns are Florence, Siena, Lucca, San Gimignano, Arezzo, Pisa etc.
Stay in Florence and take day trips to other towns.
