I'm not sure to be honest, but there aren't any huge hotels of the size that you'll find in the US / Vegas, or Tokyo Japan with 2,000+ rooms.
Most hotels upper limits are at around 550 rooms, which is considered large. As someone here says it depends on the City, but most likely Sydney has the largest hotels.
The Shangri - La ( frequently changes it's name - was formerly The Regent ) claims that it's guest rooms are the most spacious and luxurious in the city, starting from a minimum of 40 square metres .... which is HUGE !!
It has 563 guest rooms ( approx ) plusd suites which are the largest in their respective classes in Sydney, The hotel is adjacent to the Harbour overlooking the Opera House.
